Hello,
Is it possible to enable the hotkey ,for lock first monitor, at the Windows startup?
Everytime when Windows starts I must push the hotkey.

You could do this with the ExecuteHotkey script, to have the script run at startup place a shortcut to it (or the script itself) into the Start Menu > Programs > Startup folder.
Let me know if you have any questions about configuring the script.
