I have 4 24" monitors 1920x1200 and a 27" monitor 2560x1440. Because of my arrangement and that one of the monitors is in portrait, there is a gap or void of which the mouse cannot travel. I will try to illustrate my setup
Monitor 4 is the 27" Monitor 5 is a 24" in portrait V is a void where the mouse cannot travel.

So my problem is to travel my mouse between monitor 3 and 5, i must travel through 4. This would not be a problem if monitor 3 were also a 27".
So it there any setting or software that will "warp" my mouse cursor between 3 and 5 as it normally would if they were right next to each other?

I would do the following: move 2 to the left, so that 4 is aligned with 3 on the right. You can then place 5 halfway between 3 and 4, this way you can move the mouse directly to 5 from both 3 and 4.
Should look something like this:
***** *****
* 1 * * 3 * *****
***** ***** * 5 *
***** ******* * *
* 2 * * 4 * *****
***** *******
