In general hotkeys should work fine during games (this might depend on the game though), but some hotkeys won't work or won't work as expected, for example the move window, move mouse and lock mouse hotkeys.
If you want to use the lock mouse to primary monitor hotkey, you'll need to press the hotkey before launching the game, the mouse should then stay locked to the primary monitor during the game. This is useful with games which don't do this automatically.
