I would expect this to work fine, but haven't tested it myself.
I'm currently using two NVS 280 cards without problems (1x AGP + 1x PCI).

Yes they can, however a third would only install as two outputs not 3 !

This is just a limitation of Display Properties, you should be able to configure all 12 monitors with UltraMon.
See Maximum number of monitors for more on this issue.
