I am looking for something automated to replace the 'New wallpaper...' process.
Is there a way to easily define (set up), say, 100 wallpapers thru a script or a command line interface?

Not at the moment, but you could use the UltraMon Wallpaper Auto Changer 2 script to generate a new wallpaper on the fly from images from specified folders.
