Is it possible to have two profiles and have a different primary monitor in each one? Currently, it would seem that checkbox option is greyed out on my system.

Only if it's allowed by the video card driver, for example on laptops the external monitor usually can't be set as primary.
If the primary checkbox is disabled, you'll need to change the primary monitor via the settings for the video card driver (usually a tab under advanced display properties). Doing this change via UltraMon display profiles is not supported.
Please note that changing the primary monitor via the video card driver settings frequently causes the monitors to get renumbered (the external monitor becomes #1 for example), which causes problems for some UltraMon features, for example the Smart Taskbar.

Ok thanks for the quick reply.
Yes my laptop's display advanced properties lets me change the primary display, but I was hoping that Ultramon would be able to do it as well so I could effectively automate the switch using Ultramon profiles.
Basically I wanted my "home" profile to have the external monitor as the primary (since it is the bigger screen) and my "mobile" profile to have the laptop's LCD as the primary.
